Does anyone know of a way to create a floor or structural slab that is both sloped AND has a variable thickness?I can do either, but not both to the same floor. Thinking that I may need to create a generic mass.

I don't have Revit right now to create a video or some screenshots to show you but in the properties of that salb go to edit/new then in the function(layers) you must have at least 2 functions, you must turn ON the Variable of the function you want (top or bottom), then, when you are sketching the slab you have a tool to create the slope "Slope Arrow", use it, after finish the sketch when you click on the slab, in the options menu (ribbon) you will see tools for that slab, use "modify Sub-Elements"....

Check your HELP menu if you have some doubts....

I'm not sure if this actually addresses the original question.  I am going through a similar dilemma in that I need the deck (1st layer) to consistently slope across a given plane and also have the insulation be variable (2nd layer).

You can not do both functions - roof slope and varied thickness using the normal roof tools.

You can do extrusions and voids - probably by layer - to do this.
