I'm not too sure about customizing mouse clicks, but it's really easy to create keyboard shortcuts for commands. The file is usually found at C:\Program Files\Revit Structure 2010\Program\keyboardshortcuts.txt. In this file you can find the command you looking for and create a shortcut by choosing 2 or 3 letters and typing them into the left hand side in the quotations. But make sure the shortcut isn't already used by another function.
